1 5.0%孔雀綠水溶液

4.無菌檢驗用洗脫液

蛋白胨                                    10g 
氯化鈉                                    8.5g  
蒸餾水                                    1000ml
將各成分加入到 1000ml 0.03mol/L PBS液中,加熱溶解后調(diào)pH 7.27.4,于121壓力蒸汽**20min備用。

8.營養(yǎng)肉湯培養(yǎng)基

牛肉膏                                     5g

蒸餾水                                     1000ml
將各成分溶解于蒸餾水中,調(diào) pH 7.27.4,分裝,于121壓力蒸汽**20min備用。
 
9.稀釋液:胰蛋白胨生理鹽水溶液(TPS)。

氯化鈉                                           8.5g
先用900ml以上蒸餾水溶解,并調(diào)節(jié)pH值在7.0±0.2,*終用蒸餾水加至1000ml,分裝后,經(jīng)121壓力蒸汽**后使用。

5%堿性品紅乙醇溶液                       20ml

13.溴甲酚紫蛋白胨培養(yǎng)液  
蛋白胨                                   10g 
葡萄糖                                   5g
可溶性淀粉                               1g
溴甲酚紫乙醇溶液                         10ml
蒸餾水                                   1000ml
將蛋白胨、葡萄糖溶解于蒸餾水中,調(diào)pH 7.07.2,加入2% 溴甲酚紫酒精溶液,搖勻后,分裝(每管5ml),并放入一個小倒管,于 115壓力蒸汽**30min。置4冰箱備用。
 
14.嗜熱脂肪桿菌恢復(fù)瓊脂培養(yǎng)基 
蛋白胨                                    10g
牛肉膏                                    3g
可溶性淀粉                                1g
葡萄糖                                    1g
瓊脂                                      20g
蒸餾水                                    1000ml
    以上各成分蒸餾水溶解,調(diào)pH 7.07.2,裝瓶,經(jīng)115℃壓力蒸汽**30min后使用。
 
15.需氧-***瓊脂培養(yǎng)基
            酪胨(胰酶水解)                             15g
            牛肉膏                                     3g
            葡萄糖                                     5g
            氯化鈉                                     2.5g
            L-胱氨酸                                   0.5g
            硫乙醇酸鈉                                 0.5g
            酵母浸出粉                                 5g
            新鮮配制的 0.1% 刃天青溶液                 1.0ml
            (或新配制的 0.2% 亞甲藍(lán)溶液                0.5ml
                                                  0.5g~0.7g
            蒸餾水                                     1000ml
除葡萄糖和刃天青溶液外,取上述成份加入蒸餾水中,微溫溶解后,調(diào) pH 至弱堿性,煮沸、濾清,加入葡萄糖和刃天青溶液,搖勻,調(diào) pH 6.97.3,分裝于 115℃壓力蒸汽** 30min。
 
16.沙堡瓊脂培養(yǎng)基   
葡萄糖                                    40g 
蛋白胨                                    10g 
瓊脂                                      20g 
蒸餾水                                    1000ml
將上述成分混合后,加熱至完全溶解,調(diào)pH 5.6±0.2,于 115℃壓力蒸汽** 30min備用。
 
17.沙堡液體培養(yǎng)基  
葡萄糖                                    40g  
蛋白胨                                    10g 
蒸餾水                                    1000ml
將上述成分混合后,加熱至完全溶解,調(diào)pH 5.6±0.2,于 115℃壓力蒸汽** 30min備用。
 
18.無菌試驗用**培養(yǎng)基  
磷酸二氫鉀(KH2PO4)                           1g   
硫酸鎂(MgSO4·7H2O)                          0.5g

葡萄糖                                      10g
蒸餾水                                      1000ml
除葡萄糖外,上述各成分加入蒸餾水內(nèi),微溫溶解后,調(diào)節(jié)pH 6.8,煮沸,加葡萄糖溶解后,搖勻濾清,調(diào)pH 使**后為6.4±0.2,分裝,115℃壓力蒸汽**20min備用。

20.人**細(xì)胞完全培養(yǎng)基  
在人**細(xì)胞維持培養(yǎng)基中加入10%無菌小牛血清。
 
21.血瓊脂培養(yǎng)基
            營養(yǎng)瓊脂                                  100ml
            脫纖維羊血(或兔血)                         10ml
將營養(yǎng)瓊脂加熱熔化待冷至50左右,以無菌操作將10ml 脫纖維血加入后搖勻,倒平皿置冰箱備用。
 
22.霉菌培養(yǎng)基
                                          5g
             葡萄糖                                     10g
             磷酸二氫鉀                                 1g
             氯化鈉                                     5.0g
             硫酸鎂                                     0.5g
             蒸餾水                                     1000ml
    除葡萄糖外,取上述成份加入蒸餾水中,微溫溶解后,調(diào) pH 6.0 ,煮沸,加入葡萄糖溶液后,搖勻,濾清,調(diào) pH 5.66.0,分裝于 115 ℃壓力蒸汽**30min
 
23. SCDLP液體培養(yǎng)基
            酪蛋白胨                                    17g
            大豆蛋白胨                                  3g
            葡萄糖                                      2.5g
            氯化鈉                                      5g
            磷酸氫二鉀                                  2.5g
            卵磷脂                                      1g
            吐溫80                                      7g
            蒸餾水                                      1000ml
將各種成份混合(如無酪蛋白胨和大豆蛋白胨可用日本多胨代替),加熱溶解后,調(diào)pH 7.27.3,分裝于121壓力蒸汽**20min,搖勻,冷至 25℃使用。
 
24.伊紅亞甲藍(lán)培養(yǎng)基
            蛋白胨                                                                     10g
            乳糖                                                                        10g

            2%伊紅溶液                                                            2ml
            0.65%亞甲藍(lán)溶液                                                     1ml

將蛋白胨、磷酸鹽和瓊脂溶解于蒸餾水中,調(diào)pH 7.1,分裝于121壓力蒸汽**20min。臨用時,以無菌操作加入乳糖并加熱溶化瓊脂,冷至 50時,加入伊紅和亞甲藍(lán)溶液搖勻,倒平皿置4℃冰箱備用。

26. 甘露醇培養(yǎng)基
            蛋白胨                                       10g
            牛肉膏                                       5g
            氯化鈉                                       5g
            甘露醇                                       10g
            0.2%溴麝香草酚藍(lán)溶液                          12ml
            蒸餾水                                       1000ml
將蛋白胨、氯化鈉、牛肉膏加入蒸餾水中,加熱溶解,調(diào) pH 7.4,加入甘露醇和溴麝香草酚藍(lán)混勻后,分裝,于 115壓力蒸汽**20min。
 
27. 乳糖膽鹽發(fā)酵管
            蛋白胨                                       20g
            豬膽鹽(或牛,羊膽鹽)                           5g
                                                    10g
            0.04%溴甲酚紫水溶液                           25ml
            蒸餾水                                       1000ml
將蛋白胨、膽鹽及乳糖溶解于蒸餾水中,調(diào)pH 7.4,加入0.04% 溴甲酚紫水溶液,分裝(每管10ml),并放入一個小倒管,于 115壓力蒸汽**15min
 
28. 乳糖發(fā)酵管
            蛋白胨                                       20g

            0.04%溴甲酚紫水溶液                           25ml
            蒸餾水                                       1000ml
將蛋白胨及乳糖溶解于蒸餾水中,調(diào)pH 7.4,加入0.04% 溴甲酚紫水溶液,分裝(每管10ml),并放入一個小倒管,于 115壓力蒸汽**15min。

將上述成份制成溶液于121℃下**15mln,**后無菌調(diào)pH6.9±0.2備用。
 
32.注意事項
    (1)雙料乳糖膽鹽發(fā)酵管除蒸餾水外,其它成分為乳糖膽鹽發(fā)酵管的2 倍;3倍濃縮乳糖膽鹽發(fā)酵管除蒸餾水外,其它成分為乳糖膽鹽發(fā)酵管的3倍。
    (2)配制培養(yǎng)基的容器不宜用銅鍋或鐵鍋,以免影響**生長。
    (3)培養(yǎng)基用的試管口和三角燒瓶口應(yīng)用普通棉花制成的棉塞,再用牛皮紙包好。
    (4)試劑與培養(yǎng)基配制好后應(yīng)置清潔處保存,常溫下不超過1 個月。
